Effect of KfiB on heparosan synthesis in recombinant Escherichia coli BL-21

Abstract

The main objective of the project I have been working on during my internship is to determine the effects that different variants of a specific protein in the biosynthetic route of heparosan, can have on the heparosan molecule. To do so, different E. coli strain have been developed with differences in the sequence of the KfiB protein, the want of interest. Those strains have been harvested in bioreactors and induced to produce heparosan. After extraction and purification of this molecule, concentration levels in the extracellular medium part and in the pellet fraction have been determined. Moreover, heparosan molecular weight has been analysed using SEC-MALS technology.
